EDIT: Yes! I am aware that I gave them both wings, but only for the purpose that I REALLY wanted to illustrate a battle scene between them in the clouds. haha
First, I like the use of color. The background is mostly monochromatic so Twilight really stands out. It's pretty much the thing that got me to vote favorably while this was in #Best-of-MLP-FiM's voting process. Composition is also something I see as something that is totally fine and good. For the execution of this idea, it was briefly mentioned during the voting process that Twilight doesn't appear to be looking at Trixie, which does kind of take away from the believability of this piece.
IMO, anatomy might be the weakest part of this piece, but everything else I have no particular issues with. You've mostly kept with FiM style anatomy which is primarily concerned with looking cute.
I notice you've added a brow ridge to Trixie's face. IMO, unless we add a brow ridge to exaggerate a pony's anger (and squinting their eyes as well, I think that's important), it's not the best direction for adding more realism to ponies. A brow ridge is not something that would look cute (cute stuff being more baby-like and brow ridges being more prominent in adults) nor was it done in a way that makes it look more like a convincing equine.
Trixie's nose is in the wrong perspective as her nose will have no trouble looking like a side-view nose. To be fair, it's a mistake that also appears even in FiM-style.
Trixie's tail also seems to be placed oddly so it doesn't appear to be an extention of the spine, which is what a tail is.
The wings also don't appear to have an actual anatomical structure. Even FiM-style pegasus wings are loosely based off of real wings via bending a bit like real wings and having layers of feathers.
As for the anatomy on Twilight, her feathers are shaded like they have volume, but last I remember, feathers don't have very much volume if at all because they are rather flat. Also be aware of the placement of the cutie mark as I remember them being placed explicitly to their side. Beware of her hair style as Twilight's mane is not a bowlcut shape. The animators cleverly designed her mane style to not look or feel like a boring bowlcut. This style used for this piece might be a bit more realistic but it feels less like Twilight for me. Aside for also having iffy wing anatomy, that's about all that I am able to point out.
Again, anatomy is probably the only things that might be a bit lacking for this piece, but about every other aspect, as far as I've understood them, are good enough for me to vote favorably for this piece. Grats on making it past the group's cruel voting process, haha.
Ironically, it's Trixie who seem to have a worried expression on her face, while Twilight's kinda says "you're dead meat" Nice, I especially like the backgroudn and coloring... the faces are a tad "adult" for FiM, but then they do fit the concept. Good work.
